Ítem Contribucion al estudio del targua (Croton gossypifolius., Vahl): investigacion de sus propiedades antibioticoas.(1946) Rodríguez Caballero, Rafael LucasÍtem Cariotipos de las principales serpientes coral (Elapidae: Micrurus) de Costa Rica(1979) Gutiérrez, José María; Bolaños Herrera, RógerSe describen los cariotipos de las tres especies de serpientes de coral (Elapidae: Micrurus) más abundantes en Costa Rica: M. nigrocinctus, M. alleni y M. mipartitus. M. nigrocinctus presenta dos poblaciones cariotípicas que corresponden con dos de las tres subespecies descritas en nuestro país: M. n. nigrocinctus tiene un número diploide de 26 cromosomas (16 macro- y 10 microcromosomas) y M. n. mosquitensis presenta 30 cromosomas (16 macro- y 14 microcromosomas) en su juego diploide. Las hembras de ambas poblaciones cariotípicas son heteromórficas en el sexto par de cromosomas. M. alleni tiene 34 cromosomas, con 20 macro- y 14 microcromosomas en su juego dipliode. M. mipartitus también presenta 34 cromosomas, pero 14 de ellos son macro- y 20 son microcromosomas. En las tres especies se destaca la presencia de una constricción secundaria en un par de macrocromosomas.Ítem Cariotipos de diez especies de serpientes costarricenses de la familia Viperidae(1979) Gutiérrez, José María; Taylor, Richard T.; Bolaños Herrera, RógerAll the karyotypes of ten snake species of the family Viperidae from Costa Rica have a diploid number of 36 chromosomes, 16 macro- and 20 microchromosomes; females show an heteromorphism in the fourth pair. The karyological differences are restricted to the position of the centromere in the eighth chromosome pair and in the W-sex chromosome. The karyotypes of Bothrops asper, B. nigroviridis. B. schlegelii, B. nummifer, B. picadoi, B. nasutus and Crotalus durissus durissus are of an identical pattern, having a subtelocentric eighth pair and a submetacentric W-sex chromosome. The autosomal pattern of B. godmani and Lachesis muta stenophrys is similar to that of the above group, except that the W-sex chromosome is subelocentric. The karyotype pattern of B. lateralis is different from these in that it has a submetacentric eighth pair.Ítem Asociacion de algunas caracteristicas cuantitativas con el rendimiento de la soya (Glycine max (L) Merrill) en condiciones tropicales(1979) Villalobos Rodríguez, EnriqueÍtem Efectos proteolitico, hemorragico y mionecrotico de los venenos de serpientes costarricenses de los generos Bothrops, Crotalus y Lachesis(1980) Gutiérrez, José María; Chaves Mora, FernandoWe studied the proteolytic, hemorrhagic and myonecrotic activities of ten Costa Rican species of snakes from the family Viperidae. All of them showed these three activities but we did not observe any correlation between their relative magnitudes. The most myonecrotic venoms were Bothrops asper and B. schlegelii; the most hemorrhagic was B. picadoi and the strongest proteolytic activity using casein was in the venoms of B. godmani and B. lateralis. The lethality of these venoms correlated with the magnitude of tissue destruction observed microscopically. The histopathological analysis showed two kinds of necrosis: either a myolitic necrosis or a mixed necrosis, with both myolitic and coagulative areas. All the histological slides showed a strong hemorrhagic action, a polymorphonuclear leucocyte rich exudate and some vascular alterations such as angionecrosis and thrombosis.Ítem El problema de los efectos hemorrágico y mionecrótico por mordeduras de serpiente en el continente Americano(1980) Gutiérrez, José María; Bolaños Herrera, RógerLos efectos locales que aparecen como consecuencia de la mordedura de serpientes constituyen un problema de salud importante en las Américas. Sin embargo, la eficacia del suero antiofídico que se usa parece ser escasa, para neutralizarlos, aun en el caso de administrarlo inmediatamente, circunstancia que se agrava porque con frecuencia no se dispone del antiveneno hasta bastante tiempo después de ocurrido el accidente. Este estudio, describe los efectos proteolítico, hemorrágico y mionecrótico de los diversos venenos de las serpientes de Costa Rica. El cuadro hemorrágico es una característica básica de los accidentes originados por especies de la familia Viperidae, causa de la mayoría de los accidentes ofídicos de las Américas. En cuanto a la acción mioneerótica, la característica del efecto local producido por los venenos de las serpientes de la familia Viperidae es la necrosis de las células musculares, acompañada de la destrucción de tendones y cartílagos. Desde el punto de vista histopatológico, esas necrosis pueden dividirse en miolíticas y coagulativas. Respecto al tratamiento, en muchos hospitales de las Américas se utiliza como rutina la fasciotomía, aunque es indispensable utilizar antibióticos como terapia complementaria del suero antiofídico. Para resolver los problemas del tratamiento se plantean dos exigencias fundamentales, la distribución de antivenenos en gran escala y la obtención de un antiofídico más eficaz. También se ha estudiado la posibilidad de utilizar productos químicos para el tratamiento de los efectos hemorrágico y mionecrótico.Ítem Mionecrosis, hemorragia y edema inducidos por el veneno de Bothrops asper en ratón blanco(1980) Gutiérrez, José María; Arroyo Gutiérrez, Olga; Bolaños Herrera, RógerPathogenesis of the myonecrotic, hemorrhagic and edema-forming effects in white mice inoculated with Bothrops asper venom was studied at different time intervals by means of light microscopy, determination of serum levels of the enzyme creatinephosphokinase (CPK), and evaluation of edema. Although the three effects were evident in the first hour, their development was relatively independent. Myonectoric activity reached its maximum levels in the first 3 hr, as judged by serum CPK. This indicates that myonecrotic toxins have a very rapid action on muscle fibers, after which CPK levels decrease to near normal values 24 hr after venom inoculation. Histological studies showed severe myonecrosis at 3, 6, 9, 12 and 24 hr, characterized by myolytic and coagulative necrotic fibers mixed with normal fibers. Maximum hemorrhagic effect was observed between 9 and 12 hr, whereas at 6, 9, 12 and 24 hr there was an intense polymorphonuclear leucocyte infiltration. On the other hand, 1 hr after venom injection there was a local edema of 44%, reaching a maximum value of 70% within the first 24 hr, decreasing afterward. This edema is more prolonged than that induced by Agkistrodon piscivorous or Trimeresurus elegans venoms. This could have important clinical implication since local swelling contributes to tissue compression and is related to other physiological alterations. One week after venom inoculation there were fibroblasts, fragments of necrotic muscle fibers, and a mononuclear cell exudate. At this time, we did not observe hemorrhagic areas. Determination of serum levels of CPK could be a useful and powerful technique in order to quantify muscle tissue destruction due to snake bite. Since only scanty hemorrhagic and necrotic areas were observed in heart muscle, the drastic and rapid increase in CPK levels mainly reflected skeletal muscle necrosis. CPK determination is a simple and quick laboratory technique that should be used to evaluate muscle necrosis and to improve treatment in snake venom poisoning.Ítem Estudio comparativo de venenos de ejemplares recien nacidos y adultos de Bothrops asper(1980) Gutiérrez, José María; Chaves Mora, Fernando; Bolaños Herrera, RógerSe estudiaron comparativamente los venenos de ejemplares recién nacidas (menos de un mes de edad) y adultas de Bothrops asper de dos regiones costarricenses: San Carlos en la vertiente atlántica y Puriscal en la pacífica. La comparación se ba só e n la de terminación de los efectos hemorrágico, mionecrótico, edematizante, proteolítico, hemolítico y letal, así como la neutralización del efecto letal por suero antiofídico polivalente; además se efectuó una comparación bioquímica e inmunoquímica mediante electroforesis, inmunoelectroforesis e inmunodifusión en dos dimensiones. Los venenos de ejemplares recién nacidos y adultos de ambas regiones mostraron marcadas diferencias entre sí en lo que respecta a los patrones electroforético e inmunoelectroforético, aunque la inmunodifusión mostró una identidad casi completa. Los venenos de recién nacidas de ambas poblaciones son más proteolíticos, más hemorrágicos, más edematizantes y letales, en tanto que los venenos de adultos son más hemolíticos y más mionecróticos, provocando un cuadro histológico de intensa necrosis miolítica. La letalidad de los 4 venenos fue neutralizada con título similar por el suero antiofídico polivalente. Por otra parte, al comparar los venenos de especímenes adultos de ambas zonas entre sí, se apreció una ligera variación en el patrón inmunoelectroforético y una identidad total en la inmunodifusión. Los de San Carlos fueron más hemorrágicos y más mionecróticos, en tanto que los de Puriscal fueron más proteolíticos, siendo similares las magnitudes de los efectos letal, edematizante y hemolítico. The diploid number is 38 chromosomes, with 20 macrochromosomes and 18 microchromosomes. The pairs 1 and 2 are metacentrics, pair 3 is subtelocentric and pairs from 4 to 9 have the centromere in a terminal position. Estos patrones cromosómicos difieren en el número de microcromosomas y en la posición del centrómero en el octavo par de macrocromosomas. Además, dentro del grupo de M. n. nigrocinctus hubo 3 especímenes con un micro cromosoma adicional (2n = 27) y uno con dos más (2n = 28). En el grupo de especímenes de M. n. mosquitensis en dos ejemplares se observó una variación en la posición del centrómero en el cromosoma W. Some of these species are associated with flowers, rotting fruits and human cadavers.Ítem Quantitation of myonecrosis induced by myotoxin a from prairie rattlesnake (Crotalus viridis viridis) venom(1982) Ownby, Charlotte L.; Gutiérrez, José María; Colberg, Terry R.; Odell, George V.Using a pure myotoxin (myotoxin a) isolated from prairie rattlesnake Crotalus viridis viridis) venom, a comparison between two methods for quantitating myonecrosis in mice was made. Measurement of creatine kinase (CK) levels in the plasma showed two peaks in CK levels after myotoxin injection, whereas measurement by histological assay (vacuolation index) showed only one peak. The first peak in CK levels at 3 hr after injection did not correlate with a high vacuolation index, but did correlate with contraction of muscle induced by the toxin. However, there was good correlation between the two methods at 6, 12, 24, 48 and 72 hr after injection, at which time the muscle cells were necrotic. In the case of the first CK peak this method might be measuring contraction of the muscle, but the second peak was probably measuring altered permeability of the sarcolemma since vacuolation, i.e. swelling of sarcoplasmic reticulum, did follow. High plasma CK levels usually preceded high vacuolation indexes, indicating that CK release due to altered sarcolemma permeability preceded vacuolation. Electrophoretic and immunoelectrophoretic patterns showed some differences between these venoms. Venom from newborn snakes was devoid of hemorrhagic and edematigenous activities, whereas the venom from adult specimens induced these effects. On the other hand, newborn snake venom showed higher lethality and indirect hemolytic activity, and lower proteolytic activity, than venom from adult specimens. Both types of venoms induced only slight myonecrosis in mice, as judged by histological observation. The ED50 of an antivenom, in terms of absolute weight neutralized per ml of serum, was lower for the newborn specimens venom than for adult's venom, however, for each venom the number of LD50 neutralized was similar.Ítem Pathogenesis of myonecrosis induced by crude venom and a myotoxin of Bothrops asper.(1984) Ownby, Charlotte L.; Odell, George V.; Gutiérrez, José MaríaThe pathogenesis of skeletal muscle necrosis induced by crude Bothrops asper venom and isolated myotoxic phospholipase was studied using light and electron microscopy. White mice were injected intramuscularly with a dose of 2.5 micrograms/g and tissue samples were taken at 30 min and 1, 3, 6, 12, 24, and 48 hr. Toxin-injected muscle showed localized wedge-shaped lesions ("delta lesions") by 30 min, which included disrupted plasma membranes. At 1 and 3 hr the predominant type of necrotic cell contained clumped myofibrils in which individual myofilaments were indistinguishable. At later time periods there was a relaxation and redistribution of myofilaments resulting in a more homogeneous and hyaline appearance of necrotic cells. Some mitochondria were swollen and had flocculent densities, and most of them were disrupted, having only one membrane and vesiculated cristae. The basal lamina was intact at all time intervals. Phagocytosis of muscle cell debris started at 3 hr and was prominent by 24-48 hr. In crude venom-injected muscle many cells showed pathologic features identical to those observed after myotoxin injection. Crude venom also induced hemorrhage which was evident 30 min after injection, reaching its highest level by 12 hr. At 3, 6, and 12 hr some cells were undergoing different pathologic changes which appeared to be due to ischemia. Although these cells were irreversibly damaged, as indicated by ruptured plasma membrane, their myofibrillar structure was better preserved than that of toxin-affected cells. The Z line was absent, but A, I, H, and M bands were intact. As a result of Z line loss, sarcomeres were disoriented. It is proposed that the myotoxin induces myonecrosis by first altering the integrity of the plasma membrane, thereby increasing the permeability to calcium, other ions, and molecules which leads to death of the cell. Crude venom affects muscle cells in two ways: by direct action of myotoxin (s) and by ischemia due to hemorrhage.Ítem Isolation of a myotoxin from Bothrops asper venom: Partial characterization and action on skeletal muscle(1984) Ownby, Charlotte L.; Odell, George V.; Gutiérrez, José MaríaA myotoxic phospholipase has been isolated from Bothrops asper venom by ion-exchange chromatography on CM-Sephadex followed by gel filtration on Sephadex G-75. The toxin is a basic polypeptide with an estimated molecular weight of 10,700. It has both phospholipase A and indirect hemolytic activities, but is devoid of proteolytic, direct hemolytic and hemorrhagic effects. When injected i.m. into mice the toxin induces a rapid increase in plasma creatine kinase levels and a series of degenerative events in skeletal muscle which lead to myonecrosis. The toxin induces an increase in intracellular calcium levels and is able to hydrolyze muscle phospholipids in vivo. Pretreatment with the calcium antagonist verapamil failed to prevent the myotoxic activity. It is proposed that B. asper myotoxin causes cell injury by disrupting the integrity of skeletal muscle plasma membrane and that myotoxicity is at least partially due to the phospholipase A activity of the toxin.
