Aislamiento e identificación de bacterias endófitas de cultivos in vitro de tres especies tropicales para valorar la producción de metabolitos secundarios y sus posibles efectos biológicos

Resumen

La agricultura moderna se ha sustentado en el uso intensivo de fertilizantes químicos, generando impactos ambientales significativos. En este contexto, las bacterias endófitas constituyen una alternativa prometedora para reducir el uso de insumos químicos, al colonizar los tejidos internos de las plantas sanas sin causar daño y contribuir de manera importante al crecimiento y desarrollo vegetal. En este estudio se aislaron e identificaron bacterias endófitas de plantas in vitro de bambú, aguacate y vainilla, caracterizadas mediante análisis morfológicos, bioquímicos y moleculares mediante secuenciación del gen ARNr 16S. Se evaluaron rasgos funcionales vinculados a la promoción del crecimiento vegetal, como la fermentación de azúcares, la capacidad presuntiva de fijación de N2 y solubilización de fosfato, junto con la síntesis de diez fitohormonas mediante UHPLC. Adicionalmente, se verificaron los efectos biológicos de los aislados sobre semillas envejecidas de arroz y maíz, considerando parámetros germinativos y rasgos morfológicos en plántulas. En total, se obtuvieron 23 aislados bacterianos, algunos identificados a nivel de especie y otros a nivel de género. Los resultados mostraron que los aislados presentaron características promotoras de crecimiento, al presentar respuestas positivas en los rasgos funcionales evaluados y confirmar la producción de auxinas, citoquininas y giberelinas, tanto intracelularmente como en el medio circundante. En particular los aislados AB3 (Paenibacillus sp.) y AH3 (Sphingomonas echinoides) destacaron por su capacidad para favorecer la germinación y mejorar la altura y longitud de raíz en plántulas de arroz y maíz. Además, se observó que estos aislados lograron mitigar los efectos causados por el envejecimiento en las semillas, alcanzando parámetros comparables con los de semillas no envejecidas. En conclusión, los aislados obtenidos poseen potencial para modular procesos fisiológicos relevantes para el desarrollo vegetal. Los hallazgos confirman que las bacterias endófitas caracterizadas en este estudio presentan propiedades promisorias que podrían resultar de interés para sistemas agrícolas sostenibles, con potencial de favorecer el crecimiento vegetal y contribuir al desarrollo de estrategias productivas más amigables con el medio ambiente.


Modern agriculture has relied on the intensive use of chemical fertilizers, generating significant environmental impacts. In this context, endophytic bacteria represent a promising alternative to reduce the use of chemical inputs, as they colonize the internal tissues of healthy plants without causing damage and can contribute substantially to plant growth and development. In this study, endophytic bacteria associated with in vitro plants of bamboo, avocado and vanilla were isolated and identified, characterized through morphological, biochemical, and molecular analyses using 16S rRNA gene sequencing. Functional traits linked to plant growth promotion were evaluated, such as sugar fermentation, the presumptive capacity for nitrogen fixation, and phosphate solubilization, together with the quantitative synthesis of ten phytohormones using UHPLC. Additionally, the biological effects of the isolates were tested on aged rice and maize seeds, considering germination parameters and morphological traits in seedlings. A total of 23 bacterial isolates were obtained, some identified at the species level and others at the genus level. The results showed that the isolates exhibited plant growth promoting characteristics, as evidenced by positive responses in the functional traits evaluated and by the confirmed production of auxins, cytokinins, and gibberellins, both intracellularly and in the surrounding medium. Isolates AB3 (Paenibacillus sp.) and AH3 (Sphingomonas echinoides) stood out for their ability to enhance germination and improve seedling height and root length in rice and maize. Moreover, these isolates mitigated the effects of seed aging, achieving those seeds parameters comparable to those of non aged seeds. In conclusion, the isolates possess the potential to modulate key physiological processes in plant development. The findings confirm that the endophytic bacteria characterized in this study exhibit promising properties that could be of interest for sustainable agricultural system, with potential to enhance plant growth and contribute to the development of environmentally friendly production strategies.
